Sample requirements

TempO-Seq is able to efficiently quantify gene expression for a variety of samples. The assay does not require RNA extraction, purification, or cDNA synthesis and runs directly from crude cell lysates, making large screening studies of thousands to tens of thousands of samples feasible.

Apart from that, the assay has been shown to work directly from FFPE tissue up to 27 y old, and runs from with a wide variety of tissue types, including whole zebrafish larvae, tear drops, and heart biopsies, so even if a sample type is not specifically listen below, it might still be suitable for TempO-Seq analysis. Cell lysates

One of the most common applications of TempO-Seq is for high-throughput transcriptomics from cell lysates in 96 or 384-well plates, enabling efficient and affordable gene expression profiling of thousands to tens of thousands of samples. The assay runs directly from crude cell lysates, including 3D spheroids. In contrast to many other methods, TempO-Seq has an upper rather than a lower limit of cells for input. Therefore, “low cell numbers” for many other technologies are perfectly fine for use with TempO-Seq. Please get in touch to discuss your project.

FFPE tissue

Our standard FFPE assay uses between 2 mm squared to 10 mm squared from 4 - 10 µm sections, but sample inputs can be smaller (1 mm squared and below, LCM/tissue microarray, spatial transcriptomics [CellSensus]). TempO-Seq has been successfully used to carry out gene expression profiling from customer-provided laser-captured microdissected samples.

Blood

TempO-Seq runs directly from whole blood, buffy coat, and peripheral blood mononuclear cells (PBMCs). We have adapted a simple protocol for collection and storage of whole blood samples for TempO-Seq. Please get in touch to find out more.
