Introducing TempO-LINC
TempO-LINC is a novel genomics platform for single-cell gene expression profiling derived from and using the same commercial assay content as TempO-Seq™. TempO-LINC combines the benefits of the TempO-Seq assay, including the lack of a reverse transcription step, with a combinatorial split pool barcoding approach to provide transcriptome-wide gene expression measurements at single-cell resolution. TempO-LINC has a user friendly and instrument-free workflow that is highly scalable across cells and samples.

TempO-LINC has been validated on dissociated cells and nuclei across multiple species. We currently offer human, mouse, and rat whole transcriptome expression assays as well as targeted assays in other species. Please contact us to inquire about additional sample types.

Unlike traditional scRNA-Seq that requires cDNA synthesis, TempO-LINC relies on templated oligonucleotide ligation to detect target RNA. This enables the combination of high-sensitivity gene expression profiling with a robust and universal sample fixation protocol.
The targeted nature of the TempO-LINC approach enables a low percentage of counts occupied by mitochondrial and ribosomal genes. Attenuation is another unique feature of TempO-LINC that reduces the sequencing read depth occupied by specific highly expressed housekeeping genes. The net result of both is reads are more broadly distributed across genes and more total genes are detected.

For every TempO-LINC service project, we include a comprehensive and interactive sample QC report as well as single-cell gene count tables. We also include matrix formatted data directories for streamlined input into third-party software tools such as Seurat and Scanpy.

It takes 6-8 weeks for data delivery from the time that samples are submitted.

We require 50,000 cells or nuclei per sample. However, depending on the number of samples and configuration of the assay, we can accept lower numbers. We see sample capture rates of ~50% across a variety of sample types. Please contact us in advance to discuss your specific samples if you plan on submitting less than the minimum requirement.

There are no unique requirements for the preparation of single-cell or single-nucleus suspension with the TempO-LINC Fixation Kit. Prior to fixation, cells should have high viability. Cells and nuclei should also be handled in a way to minimize any impact on transcriptional signatures and prevent RNA degradation.
